چکیده

Neutrophils are multifaceted cells that, upon activation, release meshes of chromatin associated with different proteins, known as neutrophil extracellular traps (NETs). Leishmania amazonensis promastigotes and amastigotes induce NET release, we have identified the signaling pathways involved in extrusion activated by promastigotes. Amastigotes maintain infection vertebrate hosts, shown association NETs human biopsies cutaneous leishmaniasis. However, interaction neutrophils remains poorly understood. Our study aimed to characterize formation induced axenic from L. infantum, causal agent visceral Human pretreated pathway inhibitors were incubated amastigotes, was quantified culture supernatant. Amastigote viability checked after incubation NETs. We found that stimulated these requires participation elastase peptidyl arginine deaminase involvement PI3K, ROS, calcium. Moreover, not susceptible NET-mediated killing. Altogether, findings improve our comprehension implicated between neutrophils.

DNA transformation of Leishmania infantum axenic amastigotes and their use in drug screening.

Protocols for DNA electroporation in Leishmania promastigote cells are well established. More recently, in vitro culture of axenic Leishmania amastigotes became possible. We have established conditions for DNA transformation of axenically grown Leishmania infantum amastigotes. Parameters for DNA electroporation of Leishmania axenic amastigotes were systematically studied using luciferase-mediat...

Leishmania amazonensis amastigotes trigger neutrophil activation but resist neutrophil microbicidal mechanisms.

Neutrophils are the first cells to infiltrate to the site of Leishmania promastigote infection, and these cells help to reduce parasite burden shortly after infection is initiated. Several clinical reports indicate that neutrophil recruitment is sustained over the course of leishmaniasis, and amastigote-laden neutrophils have been isolated from chronically infected patients and experimentally i...

3'-nucleotidase/nuclease activity allows Leishmania parasites to escape killing by neutrophil extracellular traps.

Leishmaniasis is a widespread neglected tropical disease caused by parasites of the Leishmania genus. These parasites express the enzyme 3'-nucleotidase/nuclease (3'NT/NU), which has been described to be involved in parasite nutrition and infection. Bacteria that express nucleases escape the toxic effects of neutrophil extracellular traps (NETs). Hence, we investigated the role of 3'NT/NU in Le...

Leishmania amazonensis promastigotes induce and are killed by neutrophil extracellular traps.

Neutrophils are short-lived leukocytes that die by apoptosis, necrosis, and NETosis. Upon death by NETosis, neutrophils release fibrous traps of DNA, histones, and granule proteins named neutrophil extracellular traps (NETs), which can kill bacteria and fungi. Inoculation of the protozoan Leishmania into the mammalian skin causes local inflammation with neutrophil recruitment. Here, we investig...

Killing by neutrophil extracellular traps: fact or folklore?

Neutrophil extracellular traps (NETs) are DNA structures released by dying neutrophils and claimed to constitute a new microbicidal mechanism. Killing by NET-forming cells is ascribed to these structures because it is prevented by preincubation with DNase, which has been shown to dismantle NETs, before addition of the target microorganisms. Curiously, the possibility that the microorganisms ens...
